ABSTRACT

ObjectiveTo investigate the material basis of homologous and heterogeneous effect of Aurantii Fructus Immaturus(AFI) and Aurantii Fructus(AF) based on the total statistical moment analysis and molecular connectivity index(MCI). MethodRelevant literature at home and abroad and Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP) were consulted to establish the chemical composition database of AFI and AF, and set up their fingerprints by ultra-high performance liquid chromatography(UPLC), and the total statistical moments and similarity parameters of the fingerprint were calculated. According to MCI, all components of AFI and AF were divided into different component groups, the average values of 0-8th order(0χ-8χ) MCI of the common component groups of AFI and AF were calculated. ResultThe values of total zero-order moment(AUCT) of AFI and AF were (10.57±2.45)×106, (5.09±0.89)×106 μV·s, the values of total first-order moment(MCRTT) were (11.57±1.58), (12.10±1.29) min, the values of total second-order moments(VCRTT) were(24.49±2.30), (26.49±2.54) min2, respectively. It showed that qualitative and quantitative parameters of AFI and AF were significantly different. The components with high similarity such as neohesperidin, hesperidin and narirutin were screened as the common potential pharmacodynamic components of AFI and AF. The non-common components of AFI, such as alysifolinone and imperatorin, and the non-common components of AF, such as neoeriocitrin and isosakuranin, with high similarity were screened out as potential heterogeneous components of AFI and AF. The composition groups of AFI and AF were classified into six categories, and the similarities between the composition groups of AFI and AF and the total constituents were 0.872-0.979 and 0.918-0.997, the average values of 0χ-8χ MCI of alkaloids in AFI and AF were 3.65 and 3.14, the average values of 0χ-8χ MCI of flavonoids were 8.47 and 8.47, the average values of 0χ-8χ MCI of volatile oils were 2.71 and 3.48, respectively. It showed that there were some differences in MCI of chemical constituents(groups) between AFI and AF. ConclusionThe chemical constituents(groups) of AFI and AF not only differ in content and species, but also in structural characteristics and structure-activity relationship, which can provide a basis for further explaining the scientific connotation of homologous and heterogeneous effect of AFI and AF.

ABSTRACT

ObjectiveTo analyze the fingerprint of six pungent herbs based on the molecular connectivity index(MCI)and the matching frequency total statistical moment method, and to study the division and integration of the "imprinting template" of their volatile components, so as to find the common "imprinting template" characteristics of the pungent herbs. MethodThe volatile components of six pungent herbs were extracted by steam distillation, and their fingerprints were established by gas chromatography-mass spectrometry(GC-MS) with a programmed temperature increase(80 ℃ for 5 min, 5 ℃·min-1 to 200 ℃ for 5 min, 2 ℃·min-1 to 230 ℃ for 10 min), a splitting ratio of 20∶1, an electron bombardment ion source(EI) and the detection range of m/z 35-650, and the average MCI and total statistical moment parameters of the fingerprints were calculated. Then the matching frequency method was used to classify, integrate and confirm the chromatographic peaks of the fingerprints of six pungent herbs. ResultThe average zero order, first-order and second-order MCI values of the volatile components of Pogostemonis Herba, Artemisiae Argyi Folium, Atractylodis Rhizoma, Asari Radix et Rhizoma, Magnoliae Flos and Schizonepetae Herba were 9.02, 5.28 and 5.05, respectively. The average values of peak number, total zero-order moment, total first-order moment and total second-order moment were 60, 169×107, 22.49 min and 36.82 min2, respectively. The 20 integrated imprinting templates were obtained by the matching frequency method for the six pungent herbs, among which three were common imprinting templates with the retention times of (25.97±0.21),(26.90±0.20),(31.64±1.24) min, respectively, and the representative components were valencene,β-elemene, caryophyllin, etc. ConclusionMCI combined the matching frequency total statistical moment can divide and integrate the characteristics of imprinting templates of six pungent herbs, and find their common chromatographic imprinting characteristics, which can provide a reference for the determination of effective substances of pungent herbs.

ABSTRACT

ObjectiveTo explore the material basis for the difference in the efficacy of different parts of mulberry based on molecular connectivity index (MCI). MethodBy referring to the relevant literature at home and abroad and traditional Chinese medicine systems pharmacology database and analysis platform (TCMSP) database, the chemical composition database of mulberry-source medicinal materials was established. Venn analysis was carried out on the components among mulberry-source medicinal materials. The components in the database were divided into 10 categories, and the composition information was analyzed. According to MCI value, all components of mulberry-source medicinal materials were divided into different groups. The angle cosine method was used to calculate the MCI similarity. The average MCI values of the common component group from 0-8 orders and CI of mulberry-source medicinal materials were calculated. ResultThe components with high similarity such as (+)-cycloolivil, 1′-methoxy-2′-hydroxydihydromollugin, kuwanon, morusin and 1-deoxynojirimycin were selected as potential pharmacodynamic components. Mulberry-source medicinal materials could be divided into five component groups. The similarity between component groups and total components was 0.760-0.999, and the similarity between component groups was 0.248-0.999. In Mori Ramulus, Mori Folium, Mori Cortex and Mori Fructus, the average MCI values of their flavonoids from 0-8 orders were 4.57, 4.59, 6.41, 4.24, respectively. The average MCI values of alkaloids from 0-8 orders were 2.65, 4.55, 2.58, 2.78, respectively. The average CI values from 0-8 orders were 5.51, 5.49, 5.44 and 2.88, respectively. ConclusionIt is preliminarily concluded that there are differences in the flavonoids and pathways of hypoglycemic effects between Mori Cortex and the other three mulberry-source medicinal materials. The MCI values of alkaloids from 0-8 orders in Mori Folium and Mori Fructus were higher, but their inhibitory activity of α-glucosidase were lower than those of Mori Ramulus and Mori Cortex. The structural characteristics of the total components of Mori Fructus represented by CI were quite different from the other three mulberry-source medicinal materials.

ABSTRACT

Objective:To construct the targeting evaluation method of traditional Chinese medicine (TCM) preparations based on supramolecular Qi chromatography theory, and to study the liver targeting effect of Bupleuri Radix on Pien Tze Huang. Method:The molecular connectivity index (MCI) was used to analyze the characteristics of imprinted template and liver targeting tendency of TCM mainly attributed to liver meridian and components of Pien Tze Huang, and combined with target dynamics and total statistical moment principle, aimed at the independent action characteristics of multi-component imprinted template of TCM, a method for evaluating the targeting of TCM preparations was established. Hepatoma rats in Pien Tze Huang group, Bupleuri Radix group, Pien Tze Huang+Bupleuri Radix group and blank group were tested and verified. Result:After the average value of MCI of TCM mainly attributed to liver meridian was deducted, the MCI similarity between Pien Tze Huang group and Bupleuri Radix group was 0.376 8, Pien Tze Huang+Bupleuri Radix group and Bupleuri Radix group was 0.988 2, so it was predicted that Bupleuri Radix could enhance the liver targeting of Pien Tze Huang. A system for evaluating the targeting of TCM compounds was established, including relative total uptake efficiency (RUET), relative total concentration (RCT), relative imprinted tendency (RITT) and relative imprinted variance (RIVT). The RUET and RCT of liver were the highest in all tissues (RUET=1.88>1,RCT=2.30>1), and the corresponding values of other tissues were all <1, indicating that Pien Tze Huang combined with Bupleuri Radix could increase its distribution in liver and enhance liver targeting. Except for plasma, the RITT and RIVT of other tissues fluctuated around 1.0, indicating that targeted modification did not change imprinted tendency of Pien Tze Huang and had no significant effect on the types of components. Conclusion:Under the guidance of supramolecular Qi chromatography theory, a targeting evaluation parameter system can be established to characterize the multi-component imprinted effect of TCM preparations by MCI and total statistical moment parameters, so as to realize the evaluation of targeting of TCM preparations. The addition of Bupleuri Radix can increase the liver targeting of Pien Tze Huang.

ABSTRACT

Objective:The molecular connectivity index method and total statistical moment method were used to control the quality of traditional Chinese medicine (TCM), and the stability and consistency of volatile components of Lonicerae Japonicae Flos and Lonicerae Flos were clarified. Method:Volatile oils in Lonicerae Japonicae Flos and Lonicerae Flos from different producing areas was extracted for GC-MS determination with electron bombardment ion source, ion source temperature of 230 ℃, and detection range of m/z 35-650. Then National Institute of Standards and Technology (NIST) 05 and ChemicalBook database were used for qualitative analysis of these volatile components, the peak area normalization method was used for quantitative analysis, and the total statistical moment parameters and the zero-order, first-order, second-order, third-order molecular connectivity indexes of the components were calculated. Result:Number of peaks (RSD were 28.5%, 33.4%, respectively), total zero-order moments (RSD were 55.5%, 128.9%, respectively) and total second-order moments (RSD were 15.3%, 21.5%, respectively) of 10 batches of Lonicerae Japonicae Flos and Lonicerae Flos were unstable, indicating that the types and contents of volatile components fluctuated sharply, but the total first-order moments (RSD were 7.5%, 8.8%, respectively) and the zero-order, first-order, second-order and third-order molecular connectivity indexes (RSD ranged from 8.1% to 10.3% and 4.2% to 5.5%, respectively) were relatively stable, indicating that the overall "imprinting template" of the components was similar. Statistical analysis of each parameter found that there were no significant differences in the number of peaks, total first-order moments and zero-order, first-order, second-order, third-order molecular connectivity indexes between volatile oils from Lonicerae Japonicae Flos and Lonicerae Flos. Conclusion:Under the guidance of supramolecular gas evolution "imprinting template" theory, the molecular connectivity index method and total statistical moment parameters are used to jointly characterize the "imprinting template" of TCM components in vitro, which can control the stability and consistency of TCM quality.

ABSTRACT

The safety and effectiveness of Chinese materia medica (CMM) depend on its quality, and the modernization of CMM quality control system is an important prerequisite for the internationalization of traditional Chinese medicine (TCM). Up to now, the quality research approach of CMM is the key technology for TCM industry. In recent years, with the development of modern life science, chemistry science, analytical science and technology, the quality research level of CMM has been greatly improved, but it still stays on the two evaluation models based on chemical composition and biological effect. For single compound, because the effect is proportional to the content of components, the chemical composition can be used for quality control. However, for multi-component system of CMM, how to establish the dose-effect relationship between effects and components and find the markers reflecting their quality has become the focus of the quality control of CMM for TCM industry. Based on the theory of supramolecular “imprinting template” and “Qi-xi” put forward earlier, this paper first analyzed the characteristics of supramolecular “imprinting template” for the quality of CMM, clarified the essence of quality control of CMM, and proposed the attributes of supramolecular “imprinting template” to reflect the main effect of CMM. The control strategy of quality markers of CMM represented by core index will provide a new and more accurate characterization and calculation model for quality control of CMM.

ABSTRACT

OBJECTIVE: To study the relationship between the structural topological index and the area of cerebral infarction, and to explore the correlation between drugs with different molecular connectivity indices and their therapeutic effect. METHODS: Seventy-eight male SD rats (SPF grade) were randomly divided into four groups:drug group, model blank group, normal group and sham operation group, six rats in each group and to establish cerebral ischemia-reperfusion model. The next day, according to the five half-lives different molecular connectivity index of drugs or distilled water were given, so that blood drug concentration to reach a steady state, after the fifth half-life of drugs for anesthesia, and take out the rat′s brain after staining, fixed treatment. After photographing, the infarct area of rats was calculated by Image-Pro Plus 6 software. By t test and nonlinear regression correlation analysis using IBM SPSS Statistics 24 software the results were analyzed. RESULTS: In the drug group, the average area of cerebral infarction was 21.22%; in the model blank group, 37.23%; the ratio of reduced area to model area was 42.99%, P<0.05. There was significant difference between the two groups. In the piracetam group, 88.111 3% of the cerebral ischemia area was reduced, and the molecular connectivity index of the concentration in vivo was 190.72, 109.66, 74.91, which was significantly different from other common drugs (P<0.05). Nonlinear regression analysis showed that infarct size was correlated with molecular connectivity index, and the correlation coefficient of first order molecular connectivity index was the highest. CONCLUSION: Four groups of drugs have impaired effect on the treatment of cerebral infarction, and the molecular connectivity index of drugs has a significant correlation with the efficacy of drugs.

ABSTRACT

The molecular connectivity index was adopted to explore the characteristics of supramolecular imprinting template of herbs distributed to liver meridian, in order to provide scientific basis for traditional Chinese medicines(TCMs) distributed to liver meridian. In this paper, with "12th five-year plan" national planning textbooks Science of Traditional Chinese Medicine and Chemistry of Traditional Chinese Medicine as the blueprint, literatures and TCMSP sub-databases in TCM pharmacology of northwest science and technology university of agriculture and forestry were retrieved to collect and summarize active constituents of TCM distributed to liver meridian, and calculate the molecular connectivity index. The average molecular connectivity index of ingredients distributed to liver meridian was 9.47, which was close to flavonoid glycosides' (9.17±2.11) and terpenes (9.30±3.62). Therefore, it is inferred that template molecule of liver meridian is similar to physicochemical property of flavonoid glycosides and terpenes, which could be best matched with imprinting template of liver meridian.

ABSTRACT

To investigate the effect of the ingredient group of traditional Chinese medicine (TCM) with different "imprinted template" on the supramolecular solubilization ability of licorice, in order to lay a theoretical foundation for explaining the solubilization phenomenon of the components of TCM. Based on the independent supramolecular "imprinted template" rules, molecular connectivity index (MCI) and the correlation of n-octanol-water partition coefficient (logP) were used to indicate hydrophilic lipophilic capacity of TCM, and the extractum rate was used to indicate the solubilization effect of licoriceon single TCM herbs or compounds. The solubilization ability of licorice was evaluated based on MCI, logP and the extractum rate. According to the results, the correlation coefficient between MCI and logP for single herbs was 0.942, and that for single herb adding licorice was 0.916. The extractum rate of most herbs was increased after adding licorice. The correlation coefficient among the extractum rate as well as MCI and logP change values before and after adding licorice were respectively 0.837, 0.405. The correlation coefficient between MCI and logP for eight compounds was 0.937. Meanwhile, licorice had a solubilization effect on the remaining 7 compound except for Huangqi decoction. Therefore, licorice shows the solubilization effect through the independent supramolecular "imprinted template", so as to improve the hydrophilic lipophilic ability. There was a high positive correlation between the MCI and logP in ingredients for TCM, which could be used as important parameters to indicate the "imprinted template" feature for components of TCM. The study on the solubilizing effect of TCM with the supramolecular "imprinted template" theory was feasible, and will lay a foundation for the reform of single-herb dosage form.
